Abstract

Twenty years ago, estetrol was considered a weak estrogen, and interest in its clinical role was minimal. Its clinical application as a marker of fetal well-being was also abandoned due to high inter- and intra-individual variability in plasma levels. However, recent research has shown that estetrol has potential as a natural, orally bioavailable, selective estrogen receptor modulator. Estetrol has estrogenic agonistic effects on brain, uterus, vagina, and bone, but antagonistic effects on breast tumors in experimental conditions. This drug may have clinical applications such in hormone therapy, osteoporosis, and oral contraception. More studies should be conducted to determine its effectiveness and safety in humans.
